The two analyzers report progesterone on different scales. Time from the chart supplied for the instrument you are using; do not carry a threshold from one to the other.

Every kit also contains the instructions for use, a reagent registration card, a calibrator barcode sheet and disposable tips. Kits with controls add a quality control sheet with lot target values.
The assay package insert (Tisenc, ver 5.0, 2022) is written for serum and plasma and its printed reference intervals are not canine values. Progesterone thresholds are specific to the assay that produced them; values from a different analyzer or laboratory are not directly interchangeable — see why your analyzer’s chart doesn’t match the textbook.
Every reagent the assay needs is already sealed inside the single-test cartridge, so a run is four steps: pipette, load, touch start, print. Tisenc quotes 30 minutes to learn the routine.
  1. Register the assay and the lot
    The first time you use progesterone on the analyzer, enter the registration code from the reagent kit. Repeat for every new lot using the Reagent Registration Card in the box — calibration, QC and sample tests are only available once the lot is registered.
  2. Calibrate when required
    Run the three calibrators, reading their values from the Calibrator Barcode Sheet. Calibration is needed for each new lot, after four weeks on the same lot, when a control falls out of range, or after service. The curve is stored on the analyzer and valid for 28 days.
  3. Prepare the sample
    Spin the blood down and use serum or plasma (heparin or EDTA). Let cartridges, samples and controls reach room temperature. Mix on a vortex if needed and make sure there are no bubbles — pipette at least 100 µL into the sample well of each cartridge (the assay draws 80 µL).
  4. Load the rack
    Insert one cartridge per sample into the reagent rack (one to six), place the rack in the reaction chamber and add a fresh tip for each position. The built-in scanner reads every cartridge barcode automatically.
  5. Touch START
    Enter the sample IDs from the on-screen keyboard or a barcode scanner and start the run. Incubation, magnetic separation, three washes, substrate and reading all happen inside the instrument; progesterone completes in about 23 minutes.
  6. Read, print, dispose
    Results appear on screen in nmol/L (multiply by 0.314 for ng/mL) and print on the built-in thermal printer. Remove the rack and discard used cartridges and tips as clinical waste. The analyzer stores 100,000 results and keeps Levey–Jennings charts for your controls.

Good habits that protect the number

  • Keep kits at 2–8 °C, upright and out of sunlight; never freeze cartridges, and finish testing within 2 hours of a cartridge going on board
  • Do not mix cartridges, calibrators or controls between lots, and do not turn a cartridge upside down
  • Severely haemolysed samples are not recommended; centrifuge again if you can see fibrin or a lipid layer
  • Samples above 191 nmol/L (60 ng/mL) can be diluted 1:5 by hand and multiplied back — the analyzer does not dilute automatically
  • Wear powderless gloves; glove powder is a known cause of false results in enzyme immunoassays

Every kit ships with a package insert giving the exact volumes, timings and handling steps for that lot, and the Instrument User Manual covers the analyzer itself. The Tisenc Accre 8 brings laboratory chemiluminescence to the breeding kennel and reproduction clinic. It is a fully automated, single-test CLIA analyzer: you pipette the sample into a sealed reagent cartridge, load up to six cartridges at a time, and the instrument does the incubation, magnetic separation, washing and reading on its own.
